背景需求：

鋅（Zinc, Zn）是體內第二豐富的過渡金屬，是非常必要的催化、結構和調控離子。鋅離子（Zn2+）參與穩態、免疫反應、氧化應激、凋亡和衰老等生理過程。鋅被指出能用作突觸前神經元的一種保守型神經遞質，以及用作一種跨膜信號貫穿突觸前神經元。異常的鋅代謝與許多神經性疾病有關聯，包括阿爾茨海默病，帕金森病和癲癇（epilepsy）。熒光成像被證明是最佳的體內鋅離子監測技術。

產品描述：

Zinquin ethyl ester（CAS NO. 181530-09-6），是廣泛使用鋅離子熒光探針TSQ的類似物，也是一種細胞滲透性、基于喹諾酮結構的鋅離子（Zn2+）熒光探針，用來測定或觀察細胞內鋅離子?；罴毎麅鹊孽ッ附到庖阴セ鶊F產生Zinquin，防止其外流到穿透細胞膜。Zinquin與Zn2+在其結構的N原子處形成復合物（見圖1）。Zinquin經紫外激發并在藍色光譜范圍內發熒光（激發波長368nm，發射波長490nm）。Zinquin也能用來監測凋亡相關的胞內Zn2+流量變化。

染色流程（僅作參考）：

淋巴樣細胞（Lymphoblastoid Cells）的一般染色步驟：（不同細胞類型的染色條件有差異，有必要根據具體情況摸索最佳條件）

1.  配置Zinquin ethyl ester儲存液（1mg/ml in DMSO，相當于2.4mM）。儲存液置于-20℃保存一個月有效。

2.  用HBSS重懸細胞制備5-10×106個/ml懸液。

3.  加適量Zinquin ethyl ester儲存液（2.4mM）到懸液中按照1:1000倍稀釋制成1×染色工作液。

4.  37℃孵育30min。

5.  用HBSS清洗細胞3次，之后制成2-5×106個/ml懸液。

6.  用熒光顯微鏡或共聚焦顯微鏡檢測每個細胞的熒光強度。

Detect intracellular calcium/zinc ions: RAW264.7 cells were seeded into a 6-well plate at a density of 100 000 cells per well for 24 h. In order to determine the intracellular zinc concentration, the cells were incubated with zinc indicator (50 μL each well, 1:1000, MX4516, Shanghai Maokang Biotechnology) at 37 °C for 30 min. Then centrifugation was done at 2500 rpm for 5 min, the supernatant was removed, and resuspended in PBS, and used BD LSRFFortessaX-20 to detect.
